Objective: To explore the anti-aging effects of puerarin (the main constituent of Pueraria lobate) in miceMethods: 60 C57BL6 mice were randomly divided into five equal groups: control group (administered with physiological saline for 7 weeks), model group (subcutaneously injected with D-galactose at the dose of 150 mg /kg.d for 7 weeks), puerarin groups (administrated by intraperitoneal injection with three different doses for 4 weeks from the fourth week based on the model group). Result: 1SA-β-gal positive rate and expression of P53/P21WAF1, p16INK4a increased in model group(P<0.01). 2Interleukin-6 (IL-6) increased, superoxide dismutase (SOD) decreased in peripheral blood of model group(P<0.05). 3Administration of puerarin for 4-weeks significantly reversed the biochemical markers mentioned above in a dose-dependent manner(P<0.05). Conclusion: Puerarin has protective effects on senescent mice and Pueraria Lobata could be an anti-aging food.Key words: Puerarin, Anti-aging, D-galactose, Biochemical markers,
